Comparision of Detection efficiency of Lily Symptomless Virus in Lily Bulbs by ELISA , DBIA and Tissue Blotting Method

Abstract
This experiment was conducted to find out effective and economic indexing methods for production and maintenance of virus free lily bulbs. Dilution end points found using purified-LSV antigen revealed 15 to 30 times difference between enzyme linked immunosorbent assay (ELISA) and dot blot immunoassay (DBIA). DBIA was able to detect at lower concentration than ELISA. In the case of saps of infected tissues, dilution endpoint of DBIA was 2 to 4 times more sensitive than ELISA. Immunological efficiency analysis by tissue blotting was compared with ELISA and DBIA for detection of lily symtomless virus in infected lilies. Samples infected with high concentration were revealed positively by ELISA, tissue blotting and DBIA methods. Samples bearing asymptomatic and mild mosaic of low concentration tested negatively by ELISA, otherwise tissue blotting and DBIA revealed positive results.
